Ok, PLC is S7-1215C and you are using analog outputs built into the PLC with the default configuration of the AO's set as 0.20mA and QW64 and QW66.

When you look at the tag currently configured as QW64 (from PLC tag table), what value is displayed in your watch table?

You realize where the AO terminal block and terminals are, correct?  They are tricky for some people and they get the front/back set of terminals reversed in their heads and wire up the wrong terminal block.

without connect the register, always showing 2.09mA & after connecting the 250ohm register across the terminal 2M & channel 0, it is showing 0.38mA as per 27648 values.... both analog outputs have been checked but found the same result... so can you help me please?

Then it is a wiring problem, or you have somehow managed to damage the AO's, which is not likely.  It is usually pretty easy to detect everything is working properly.

Might I suggest running the AO terminals to external terminals.  The AO COMMON would have 3 terminals jumpered together.  1 to chassis ground, 1 for AO0, and the other for AO1. Then you jumper the 500Ω resistors from common and signal terminals making it easy to measure voltage.  Once the voltage output across the resistors are correct, you know the AO's are functional.

In the watch table QW64 is showing  zero as per 0 command & showing 27648 as per 100 command while in the Norm X range 0-100
