this is entirely up to you, as long as you make sure that the S5 Timer is supplied with an S5 Time format compliant value. have a look at the FAQ below which will show you ways in case you use a Siemens HMI: How do you specify the runtime for a timer via an operator panel, or how do you output the remaining runtime? Should you use a non Siemens HMi/SCADA, do a search on the subject of HMI and S5 Timer @ https://support.automation.siemens.com and you'll find plenty of other ways to accomplish this.
