The answer to your question is: yes, you can use Ethernet to download, upload, and program the S7-300 and S7-400 PLCs, if you have the correct interface on your system. - If your CPU has an RJ45 connector, connecting to an integrated Profinet port (the CPU part number will contain a -PN identificator), then you can use an Ethernet connetion between Step 7 and the CPU, with the PC's Ethernet cards. - If your S7-300 has a CP 343-1, or your S7-400 has a CP 443-1, even if he CPU does not have an integrated Profinet port, you can setup an Ethernet connecton between Step 7 and your CPU, using your computer's Ethernet card, by connecting to the CP.
