I want to send the data available in the DB to two separate controllers via Modbus communication using (MB_SERVER). Which method should I follow to send the Modbus data?

I have two CP hardware communication modules.

PLC used: CPU 1510SP-1 PN - ET200SP
CP modules : CP 1542SP-1 IRC - 2 nos

your data exchange expectation looks to me little bit strange but I try answer to you as your topic name shows. You need assign to your 2 different Server blocks each ETH port you want to use. You will have then 2 different Servers, each use own ETH port and then its up to Clients what they will do.

Ok, for sure, your problem description says to me that you want send data to 2 different Clients. Pay attention to one thing, also from your description is clear that same data source usage is expected here so in case that Clients want to write something, some signal collission can occur. Again, it is not clear to me what exactly you want to do.
