Yes, without SD and with an empty internal eMMC, the device will not boot anything. BUT: It may get beyond the point where your previous boot with SD card plugged got stuck. Are you sure that there is really nothing on the serial console in that case? When does the red LED turn on? Immediately after power on or rather after a longer while?

I'm suspecting that the firmware gets stuck accessing the SD card, and that in very early boot (it tries to look for secure boot setup instructions on the SD card). Therefore the test to start without it. Such an issue might be specific to the plugged card, thus trying to use a different one would be worth to try as well.
