You can change and monitor data using Variable Table (i.e. Vat Table in Siemens Terminology).

You can add Vat table in PLC, and best part is you don't have to download them.

 if you want to change initial value permanently you can go to DB set required values in initial value area then download DB again ,

for temp changes set actual value in that area of DB

 You can change the software online, but some precautions have to be taken in order to avoid your PLC going into stop mode.

• Make sure that you have downloaded error handling OBs. 

  Which organization blocks do you need for error handling in STEP 7 V5.5?

• Make sure that you download Datablocks before you download the functions that call them.
• Make sure you don't download hardware configuration (in this case a PLC stop is inevitable except if you have redundant system or CIR capability)
• Modifications to HMI will not force your system to go to stop mode.

But what's your target?

Is it really not to enter the CPU into stop mode and stop production? 

I  can imagine that it may be a requirement from production department.  By the way,  it doesn't related to pharmaceutical industry. All industries and all production management don't allow us to make stoppage time.

But the most important thing specially in pharmaceutical industry is the product quality. It should not be affected by your modifications. Otherwise you have to ask them to plan a stoppage time to be sure that no batch will be affected.

First you should state the used PLC technology whether it is S7 300 or S7 400 or others and the configuration your whole system.

Also you shall define you are using Ram or Flash EPROM for your CPUs

           Depends on the storage location of the values you can change them.

       1.  For M-bits or DB values   change from   vat table/ watch table.

       2. For changing I/O    change from     Force table.
