16# 02:4427 indicates that an active online connection from TIA Portal, etc was not terminated correctly or was abruptly interrupted. This could be caused by power save related settings on PC/PG, or other interruptions to the online connection.

You can do a factory reset to PLC and download the project again.

you don´t need a factory reset for that., occupied connections will be reset just by power off/on. The actual FW also has a function called Keep-Alive-connection monitoring, keepalives are sent for connections via TCP/IP. This ensures that connections are terminated if one of the communication partners fails and that connection resources are released again.

furthermore I think this has another reason. When you are able to connect over online devices, you are also able to connect with project. I would rather check if you are in the rigth IP-range with your Programming device. Also try it over extended online connection and directly to the IP, the CPU has, as shown in this FAQ:
