I guess your BackUp file has been made by using WINCC Flexible you can restore your backup by using WINCC flexible then you have to use the MIGRATION tool of the tia portal

to open the WINCC pages in TIA portal

 The backup file was generated by the Comfort Panel and extracted via USB. The project was initially made in WinCC TIA Portal. Would WinCC Flexible still be able to generate the project file from the run time file?

I'm sorry, but you can't open a WinCC TIA Portal project (or backup file) in WinCC flexible.

You can not retrive the source code from runtime file.

You need original project backup then only you can edit and download the project again in comfort panel

One more option, 

Upload the project backup thru comfort panel via pro save . But this project backup is not editable format. You can downlaod this project in same type of comfort panel only.
