Yes , it is possible to change the back up batteries while the PS is ON.Follow the instructions in "7.1 Replacing the Backup Battery" of the following manual:

Hello Gaston; From a note in the S7-400 module data manual, discussing the battery option on the power supply mdules (both AC and DC), it seems they recommend having the power supply installed and powered when installing thew batteries. Hello Dear Gaston_Sanchez As long as the power supply module is under load (Is On), you can replace the batteries The attached file provides comprehensive information on how to install and use the battery.
