Yes , it is possible to change the back up batteries while the PS is ON.Follow the instructions in "7.1 Replacing the Backup Battery" of the following manual: Automation System S7-400 Hardware and Installation

Hello Dear Gaston_Sanchez As long as the power supply module is under load (Is On), you can replace the batteries The attached file provides comprehensive information on how to install and use the battery.
